I know mono line wears down and needs to be replaced periodically. But what about mono on a spare spool thay has not been uses since I put the line on 2 years ago?

1) Depends on where you kept it. A cool, dark place? A warm / hot place perhaps exposed to sunlight? If the former, I'd give it a try. If the latter, strip it and replace.
2) A bigger problem might be the memory the line has developed sitting on the spool for that length of time. A PITA to deal with.
3) Hey, mono is cheap. Treat yourself, especially if you're going to be doing some serious fishing.

I agree, if it hasn't been in the sunlight and hasn't been exposed to heat, it should still be good.
