Having never trolled before, decided to buy a couple of Bass Pro's Gold Cup Line Counters. Bought some Berkley Fire Line and fed it on to the reel. Tested the Line Counter by measuring it against a 15 foot mark only to find out it counted 20 feet. Having said that, I realized that the Berkley spool only contained 125 yds and this reel can hold 280 yards. Because I didn't have a full spool, would this attribute to the difference in measurements?

Yup. It should be calibrated using a full spool.
Not all line counters are right on but at least it gives you the info to repeat what you are doing.
You will need to put some backing on the reel. Any kind of mono will do. I use anything over 10lbs for backer so that I dont need as much. Since you bought two reels just leave the fireline on one reel then tie on the backing using a uni to uni knot and fill er up with the mono. Now, tie the mono to the 2nd reel and transfer it back to the 2nd reel. Keep track of the line counter to see how much mono you need and put it on reel #1 and top off with your fireline.
